(RTTNews) – It’s been only a few days since the consumer health division of Swiss drug maker Novartis AG (NVS: News ) temporarily suspended operations at Lincoln, Nebraska, production facility and recalled certain over-the-counter drugs – Bufferin, Excedrin, Gas-X Prevention and NoDoz, due to mix-up of products. Now, the animal health division of Novartis has warned that a wrong tablet may be found in bottles of Clomicalm, which is prescribed to treat behavioral disorders in dogs.

Novartis Consumer Health Inc., the consumer health division of Novartis, initiated the voluntary recall of the OTC drugs on January 8, following consumer complaints of chipped and broken pills in the products and inconsistent bottle packaging line clearance practices.

The opiate products manufactured and packaged for Endo Pharmaceuticals Holdings Inc. (ENDP) by Novartis Consumer Health at the Lincoln, Nebraska manufacturing site have also been put on alert for a potential product mix-up.

However, there have been no reports of related adverse events as a result of these issues.

On January 25, Novartis Animal Health US, Inc. warned that due to potential packaging issues at its manufacturing facility, there is a rare possibility that a wrong tablet may be found in bottles of Clomicalm. The animal health unit of Novartis said that it has not received any reports where a patient experienced a product mix-up, or any reports of adverse events attributable to a product mix-up.

As a precautionary measure, Novartis is recommending that before dispensing Clomicalm, the contents of the bottle should be examined for tablets that are broken or incorrect in color, shape or size.

On January 10, Novartis had already warned that the temporary suspension of operations at the Lincoln, Nebraska, production facility would affect the availability of some Novartis Animal Health products for the immediate future. The manufacturing plant produces both OTC and animal health products.

The animal health products affected include, Interceptor Flavor Tabs, Sentinel Flavor Tabs, Clomicalm, Program Tablets and Suspension, and Milbemite. Production of Deramaxx, which was recently moved to the site, is also affected, although supplies on hand will continue to be shipped, according to the letter sent by Novartis to the veterinary community.

The Texas Veterinary Cancer Registry aims to advance veterinary cancer research and to create connections between researchers, veterinarians and owners of pets with naturally-occurring disease that could provide the critical data to someday help eliminate cancer as we know it.

By including your pet in this database of animals with cancer we will gain important information regarding common cancers in our pet populations. Such information will be useful for applying new treatments to dogs and cats and for better understanding the progression of these life-threatening diseases. Improved understanding of the similarities and differences between cancer in our pets and cancer in humans may help improve treatment in both humans and pets.

How The Registry Works

By working to improve cancer treatments for animals, the Texas Veterinary Cancer Registry will also advance the fight against the disease in humans.

Since many diseases that affect both animals and humans are nearly identical, studying the disease in animals helps scientists predict how the disease or its treatment will affect humans. Also, NOAM diseases typically progress faster in animals than humans because of their shorter life-spans. Studying drugs and devices in animal models may significantly decrease the cost of development by identifying relevant safety issues, effectiveness, biomarkers, and genetic markers prior to expensive human clinical trials. Additionally, development of databases of companion animal diseases will provide an important and unique source of information that can be utilized to determine disease trends.

Step 1: Create a Network of Veterinary Oncologists

The Texas Veterinary Oncology Group is a team of veterinarian oncologists statewide working to advance the study of drug therapies for dogs and humans. Through this statewide network of experts, the Texas Veterinary Cancer Registry connects animals that have developed naturally-occurring cancers with researchers and clinicians advancing breakthrough treatments.

Step 2: Collect Data for the Registry

The TVCR will collect, compile, and analyze crucial information from pet-owners and veterinarians about pets that have been diagnosed with various forms of naturally-occurring diseases (most often, cancer). This information will be used to advance the care and treatment of animals with cancers with the hope of eventually matching animals with relevant clinical studies. Our ultimate goal is to enhance the treatment of both human and veterinary cancers through the collection and dissemination of new information regarding cancer therapies.

The registry will allow owners and veterinarians to share detailed information about their pet’s disease in an anonymous and confidential fashion. It will also allow pet owners to connect with other people whose pets have similar diagnoses and discuss treatment options and outcomes. Through this connection, pets will benefit from leading edge treatments and a better quality of life.

Step 3: Applying the Database

The TVCR will review the database for potential candidates for clinical trials that aim to find treatments for specific NOAM diseases.

To enter your pet or patient into the database for consideration, please register your pet.

Top 10 Most Frequently Reported Poison Dangers for Dogs in 2011 reported by Pet Poison Helpline at 800-213-6680

The veterinarians at Pet Poison Helplinehave perused their records for 2011, and determined the “Top Ten List” of potential poisons in our homes and yards that were the most commonly reported during 2011.

“Each year we examine our records to determine what contributed to the most calls from pet owners and veterinarians,” said Justine Lee, DVM, DACVECC and associate director of veterinary services for Pet Poison Helpline. “Since we spent the most time diagnosing and specifying treatments for dog-related emergencies, we’ve broken them down and produced a ‘top ten list’ designed to educate dog owners and provide veterinarians with the latest facts and statistics.”

Below is the Top Ten List from Pet Poison Helpline. Items are presented in order of frequency starting with foods, which accounted for the highest number of poisoning cases in 2011.

1. Foods – specifically chocolate, xylitol, and grapes/raisins.

Certain types of chocolate are very toxic to dogs. The chemical causing toxicity in chocolate is theobromine (a relative of caffeine). The darker, more bitter, and more concentrated the chocolate is, the more dangerous it is. Many sugarless gums and candies contain xylitol, a sweetener that is dangerous to dogs. When ingested, even in small amounts, it can result in a life-threatening drop in blood sugar or even liver failure.Raisins and grapes are often overlooked as one of the most toxic foods to dogs, and can result in kidney failure.

2. Insecticides – including sprays, bait stations, and spot on flea/tick treatments.

Ingestion of insecticides and pesticides, especially those that contain organophosphates (e.g., disulfoton, often found in rose-care products), can be life-threatening to dogs, even when ingested in small amounts. While spot-on flea and tick treatments work well for dogs, they can be very toxic to cats when not applied appropriately. Cat owners should read labels carefully, as those that contain pyrethrins or pyrethroids (a derivative of the Chrysanthemum flower), are severely toxic if directly applied or ingested.

3. Mouse and rat poison – rodenticides.

There are many types of chemicals in mouse and rat poisons, all with different active ingredients and types of action, making all of them potentially poisonous to dogs. Depending on what type was ingested, poisoning can result in internal bleeding, brain swelling, kidney failure, or even severe vomiting and bloat. Mouse and rat poisons also pose the potential for relay toxicity, meaning pets – and even wildlife – can be poisoned by eating dead rodents poisoned by rodenticides.

4. NSAIDS human drugs – such as ibuprofen, naproxen.

Common drugs including NSAIDs (e.g. Advil®, Aleve® and Motrin) can cause serious harm to dogs when ingested, causes stomach and intestinal ulcers as well as potential kidney failure. The use of human NSAIDs in dogs is dangerous and should never be given without consulting Pet Poison Helpline or a veterinarian.

5. Household cleaners – sprays, detergents, polishes.

Strong acidic or alkaline cleaners pose the highest risk due to their corrosive nature, and include common household products like toilet bowel cleaners, lye, drain cleaners, rust removers, and calcium/lime removers. Remember that “natural” does not necessarily mean safe, as some natural products can cause severe reactions. While general cleaners like glass products, spot removers and most surface cleaners have a wide margin of safety, it is still wise to keep them out of reach.

6. Antidepressant human drugs – such as Prozac, Paxil, Celexa and Effexor.

Of all prescription medications, antidepressants account for the highest number of calls to Pet Poison Helpline. When ingested, they can cause neurological problems in dogs like sedation, incoordination, agitation, tremors and seizures.

7. Fertilizers – including bone meal, blood meal and iron-based products.

While some fertilizers are fairly safe, certain organic products that contain blood meal, bone meal, feather meal and iron may be especially tasty – and dangerous – to dogs. Large ingestions can cause severe pancreatitis or even form a concretion in the stomach, obstructing the gastrointestinal tract.

8. Acetaminophen human drugs – such as Tylenol and cough/cold medications.

Sizeable ingestions of acetaminophen can lead to severe liver failure and even dry eye in dogs. However, it should be noted that it is a more significant threat to cats, as a single Tylenol tablet can be fatal.

9. Amphetamine human drugs – ADD/ADHD medications like Adderall and Concerta.

Medications used to treat ADD (Attention Deficit Disorder) and ADHD (Attention Deficit Hyperactivity Disorder) contain potent stimulants, such as amphetamines and methylphenidate. Even minimal ingestions by dogs can cause life-threatening tremors, seizures, elevated body temperatures and heart problems.

10. Veterinary pain relievers – specifically COX-2 inhibitors like Rimadyl, Dermaxx and Previcox.

Carprofen, more commonly known by its trade name Rimadyl, is a veterinary-specific, non-steroidal anti-inflammatory drug. While it is commonly used for osteoarthritis, inflammation, and pain control in dogs, if over-ingested in large amounts, it can result in severe gastric ulceration and acute kidney failure in dogs.

PHILADELPHIA — Members of a University of Pennsylvania research team have shown that they can prevent, or even reverse, a blinding retinal disease, X-linked Retinitis Pigmentosa, or XLRP, in dogs.

The disease in humans and dogs is caused by defects in the RPGR gene and results in early, severe and progressive vision loss. It is one of the most common inherited forms of retinal degeneration in man.

“Every single abnormal feature that defines the disease in the dogs was corrected following treatment,” said lead author William Beltran, assistant professor of ophthalmology at Penn’s School of Veterinary Medicine.

“We were thrilled,” said senior author Gustavo Aguirre, professor of medical genetics and ophthalmology at Penn Vet. “The treated cells were completely normal, and this effect resulted from introducing the normal version of the human gene into the diseased photoreceptor cells.”

The similarities between humans and dogs, in terms of both eye anatomy, physiology, disease characteristics and positive response to this gene therapy, raise hope for a clear path to human therapies.

Beltran and Aguirre collaborated with Artur Cideciyan and Samuel Jacobson at the Scheie Eye Institute, part of the University of Pennsylvania’s Perelman School of Medicine. This achievement results from more than 10 years of close collaboration between the scientists at Penn’s veterinary and medical Schools and the University of Florida.

In addition to others at Penn Vet, Scheie and Florida, researchers at the universities of Michigan and Massachusetts and the National Eye Institute at the National Institutes of Health contributed to the research.

The study will be published in the journal Proceedings of the National Academy of Sciences.

The gene therapy approach used takes advantage of a viral vector — a genetically modified virus that doesn’t cause disease and is unable to divide — to deliver the therapeutic RPGR gene specifically to diseased rods and cones. In the absence of treatment, these cells malfunction and progressively die.

The research team has previously successfully applied a similar approach to two other heritable vision disorders that occur in both humans and dogs: Leber congenital amaurosis and achromatopsia. The present study was more challenging, as it was necessary to target both main classes of photoreceptor cells.

While the exact disease mechanism of the RPGR form of XLRP is still unknown, the researchers were able to successfully treat dogs with two different RPGR mutations. The mutations disrupt photoreceptors in different ways, but both ultimately cause them to become useless for vision. While this form of blindness is rare in dogs, it is common in humans. Patients with XLRP usually begin to lose night vision as children and become almost totally blind by middle age.

This is the first proof that this condition is treatable in an animal model; a single subretinal injection administered to the diseased dogs led to functional and structural recovery. The dogs’ recovery was assessed using a variety of methods that are used clinically in patients, such as electroretinography and optical coherence tomography.

The researchers feel the results are promising and relevant for translation to the clinic.

“We are intervening to treat both classes of photoreceptor cells, rods and cones, and that has never been done before in a large animal model,” Beltran said. “And not only can we prevent the disease onset but also restore the remaining photoreceptor cells to normal once the disease is ongoing.”

While the ability to repair both rods and cones was itself a first, the research team went further, showing that its treatment also repaired the photoreceptor connections to other retinal neurons that eventually send visual signals to the brain, another first.

“This not only provides hope for reversing XLRP but potentially for any form of photoreceptor degeneration,” Aguirre said. “Altered inner retinal wiring is a common feature for these diseases that has been considered irreversible.

“The study required a combination of genetic tools and surgical technique to make sure the therapy targeted only the diseased cells. The viral vector had to be injected in the sub-retinal space so as to be in close proximity to the photoreceptors. Likewise, you need to ultimately deliver the therapy to the right location of the retina,” Aguirre said.

“In the human disease, careful characterization of the areas of the retina that need to be treated is going to be critical for therapy to succeed in the clinic,” Cideciyan said.

The genetic aspect of the viral vector used in this study involved a double safeguard. The first safety feature was to use a viral vector that is known to predominantly target both rods and cones but not other cells. The second safeguard involved attaching the healthy RPGR gene to a “promoter,” a piece of genetic code that would “switch on” the gene only if the virus penetrated the correct cell.

Selecting the right promoter was critical; the lead researchers at the University of Florida, William W. Hauswirth and Alfred S. Lewin, had to find one that that would be turned on exclusively in rods and cones. This way, even if the virus made its way to a non-photoreceptor cell, that cell would not start activating the RPGR gene.

That both the promoter and the RPGR gene it activates are taken from humans is a strong sign that the treatment may be translatable to patients.

“While there is still much work to do to assess long-term efficiency and safety with this approach, there is hope that this vector and knowledge could be used in a few years to treat the many patients losing vision from XLRP,” Jacobson said.

In addition to Beltran, Aguirre, Cideciyan, Jacobson and Hauswirth, the research was conducted by Sem Genini and Simone Iwabe of Penn Vet; Alejandro J. Román, Malgorzata Swider, Alexander Sumaroka and Tomas S. Alemán of Scheie; Alfred S. Lewin, Diego S. Fajardo, Vince A. Chiodo, Wen-Tao Deng and Sanford L. Boye of Florida; Hemant Khanna of Michigan and Massachusetts; and Anand Swaroop of Michigan and the National Eye Institute.

The research was supported by the National Institutes of Health, the Foundation Fighting Blindness, a Fight for Sight Nowak family grant, the Midwest Eye Banks and Transplantation Center, the Macula Vision Research Foundation, the Van Sloun Fund for Canine Genetic Research, Hope for Vision, and Research to Prevent Blindness.

Researchers discover novel anti-viral immune pathway in the mosquito | Virginia Tech News | Virginia Tech.

BLACKSBURG, Va., Jan. 10, 2012 – As mosquito-borne viral diseases like West Nile fever, dengue fever, and chikungunya fever spread rapidly around the globe, scientists at Virginia Tech are working to understand the mosquito’s immune system and how the viral pathogens that cause these diseases are able to overcome it to be transmitted to human and animal hosts.

In nearly every part of the world, humans and animals experience high levels of morbidity and mortality after being bitten by mosquitoes infected with viruses. More than 100 different viruses transmitted by blood feeding arthropods like mosquitoes have been associated with human or animal disease.

Two especially prolific vectors are the yellow fever mosquito (Aedes aegypti) and Asian tiger mosquito (Aedes albopictus), which is easy to spot because of its striped patterning. Although native to Africa and Asia, these insects can spread through the western world by hitching rides in used tires, which trap water to create a perfect breeding site.

Virginia Tech researchers recently identified a novel anti-viral pathway in the immune system of culicine mosquitoes, the insect family to which both vectors belong. Kevin Myles and Zach Adelman, both associate professors of entomology in the College of Agriculture and Life Sciences, publish their findings this month in PLoS Pathogens.

“We have previously shown that an antiviral response directed by small interfering RNAs (siRNAs) is present in culicine mosquito vectors. However, we show here that another class of virus-derived small RNAS, exhibiting many similarities with ping-pong-dependent piwi-interacting RNAs (piRNAs) is also produced in the soma of culicine mosquitoes,” they explain. Myles, Adelman and co-workers made use of a technique called next generation sequencing to aid in their discovery.

The newly discovered antiviral pathway appears to act redundantly to the previously described siRNA pathway, indicating a robust immune system, said Myles. Thus, understanding how viruses get around the mosquito’s dual antiviral responses poses an increasingly interesting challenge to scientists.

“In the case of mosquito-borne pathogens, our health depends as much on the mosquito’s immune response as it does on our own immune response, yet surprisingly little is known about the immune system of the mosquito,” Myles said.

After coming to Virginia Tech in 2005, Myles and Adelman built a research program devoted to work in this sub-field. They are members of Virginia Tech’s Vector-Borne Disease Research Group and affiliated faculty members in the Fralin Life Science Institute. The mission of the Vector-Borne Disease Research Group is to elucidate the fundamental mechanisms involved in the transmission and pathogenesis of vector-borne infectious organisms, to deepen understanding of the nature of infectious disease, and to lead the search for novel approaches to disease mitigation.

The Fralin Life Science Institute strategically invests in targeted research areas within the life sciences. Such investments include recruitment and set-up support for new faculty members, retention and recognition of established faculty members, seed funds for new research projects, equipment purchases, graduate student recruitment and support, undergraduate research support, and support for outreach activities. Research initiatives within the life sciences receiving the highest priority for support include vector-borne disease, infectious disease, plant sciences, ecology and organismal biology, obesity, and cancer biology. The Fralin Life Science Institute is also actively engaged in cooperative partnerships with colleges, departments, and other institutes that support the life science community.

Seven Virginia Tech institutes support research and creative scholarship in strategically important areas, drawing upon the university’s established strengths. The Virginia Tech Carilion School of Medicine and Research Institute, the Virginia Tech Transportation Institute, and the Virginia Bioinformatics Institute focus on national research priorities, including translational health and medical research, national security, and safe infrastructure. The Institute for Critical Technology and Applied Science, Fralin Life Science Institute, and the Institute for Society, Culture, and Environment provide organizational and financial support to develop opportunities at the intersection of engineering, science, and medicine; target infectious disease; and advance human development. The Institute for Creativity, Arts, and Technology promotes creativity, critical thinking, and life-long learning.

Helminths induce potent T helper 2 (TH2)-type immune responses that can mediate worm expulsion, but the role of this response in controlling the acute tissue damage caused by migrating multicellular parasites through vital tissues remains uncertain. We used a helminth infection model in which parasitic nematode larvae migrate transiently through the lung, resulting in hemorrhage and inflammation. We found that IL-17 initially contributed to inflammation and lung damage, whereas subsequent IL-4 receptor (IL-4R) signaling reduced elevations in IL-17 mRNA levels, enhanced the expression of insulin-like growth factor 1 (IGF-1) and IL-10 and stimulated the development of M2 macrophages, all of which contributed to the rapid resolution of tissue damage. These studies indicate an essential role for TH2-type immune responses in mediating acute wound healing during helminth infection.
